Single Copper Atom Photocatalyst Powers an Integrated Catalytic Cascade for Drug-Resistant Bacteria Elimination

Abstract: To address the issue posed by drug-resistant bacteria and inspired by natural antimicrobial enzymes, we report the atomically doped copper on guanine-derived nanosheets (G–Cu) that possess the integrated catalytic cascade property of glucose oxidase and peroxidase, yielding free radicals to eliminate drug-resistant bacteria upon light irradiation. Density functional theory calculations demonstrate that copper could notably promote oxygen activation and H2O2 splitting on the G–Cu complexes. Further all-atom sim… Show more

“…Subsequently, we studied the bactericidal efficacy of Lipid-CIP nanoassemblies against a representative drug-resistant bacterial stain, Staphylococcus aureus ( S. aureus ) Xen36. Pristine Lipid nanoassemblies (without the drug loading) showed negligible bacteriostatic and bactericidal efficacy, with minimum inhibitory concentration (MIC) and bactericidal concentration (MBC) higher than 250 μg/mL (Figure S29, Table S3, Supporting Information). The MIC and MBC values of CIP in Lipid-CIP nanoassemblies were 4-fold lower than free CIP and 2-fold lower than CIP-PBA.…”

“…With all the aDCNs in hand, we then studied the bactericidal efficacy of the aDCNs, as summarized in Table S3 . Undoubtedly, PVP, B and C domains demonstrated negligible antimicrobial efficacy with MIC and MBC values higher than 500 μg/mL against the clinically isolated Escherichia coli ( E. coli ) and multi-drug resistant Staphylococcus aureus ( S. aureus Xen36) [ 43 , 49 ]. The clinically isolated E. coli is a polymyxin-resistant strain with a MIC value of 32 μg/mL.…”

“…The GOx-assisted cascade reaction process of the CPNC, CPNC@GOx, and CPNC@GOx-Fe 2+ in the presence of glucose was verified by the oxidation of 3,3″,5,5″-tetramethylbenzidine (TMB) that reacts with •OH to yield blue oxTMB with a characteristic UV absorption peak at 652 nm (Figure 2g; Figures S13b and S14c, Supporting Information). [49,50] CPNC@GOx-Fe 2+ alone exhibited rapid catalytic activity toward the oxidation of TMB, resulting in a visible blue coloration (Figure S13b, Supporting Information). However, due to the peroxidase-like catalytic activity of CPNC (Figure S14a, Supporting Information), CPNC@GOx can also undergo a GOx-assisted cascade reaction process.…”
