Single-lipid dynamics in phase-separated supported lipid bilayers

Abstract: Phase separation is a fundamental organizing mechanism on cellular membranes. Lipid phases have complex dependencies on the membrane composition, curvature, tension, and temperature. Single-molecule diffusion measures a key characteristic of membrane behavior and relates to the effective membrane viscosity. Lipid diffusion rates vary by up to ten-fold between liquid-disordered (Ld) and liquid-ordered (Lo) phases depending on the membrane composition, measurement technique, and the surrounding environment. “…Transmembrane receptors on the surface of cells typically cluster from the nanometer to the micrometer scale to initiate signal transduction [ 43 , 44 ]. Phase separation is a critical cellular mechanism of activity on biological membranes [ 45 ]. Transitions between ordered and disordered phases are feasible in lipid bilayer model membranes, and phase separations can occur in membranes comprised of multiple lipid species [ 46 ].…”

“…There is still no complete consensus on the interpretation of electronic microphotographs obtained using this method. Shestrand [31] believes that under normal conditions, there is a kind of fusion of two membrane elements.…”
