Single-Molecule Observations of Surfactant Diffusion at the Solution−Solid Interface

Abstract: Individual fatty acid molecules adsorbed at the interface between hexadecane and fused silica have been tracked using total internal reflection fluorescence microscopy. Two cooperative diffusive mechanisms are observed: continuous small-scale Brownian motion and occasional large "jumps." The continuous diffusion exhibits evidence of confinement. The effective interfacial diffusion coefficients for each mechanism increase systematically with temperature; an Arrhenius analysis gives an activation barrier of appr… Show more

“…Total internal reflection fluorescence microscopy (TIRFM) [41] was used in conjunction with intermolecular Förster resonance energy transfer (FRET) and alternating laser excitation, allowing direct quantitative analysis of molecular search behavior as a function of initial searcher-target distance [42,43] These search times were directly compared to theoretical predictions based on Brownian searching and intermittent desorption-mediated searching respectively [15].…”

“…The diffusive trajectories of individual fluorescent molecules were determined using custom algorithms implemented in Mathematica (Wolfram Research, Champaign, IL) as described in Honciuc et al (35). Any trajectory with a total residence time shorter than 0.75 s was ignored for the purpose of statistical analysis.…”

“…In previous work, we demonstrated the ability of single-molecule total internal reflection fluorescence microscopy (TIRFM) to quantify the adsorption 18,19 and diffusion [19][20][21] of surfactant probes at the solid-liquid interface in an effort to understand fundamental dynamic interfacial behaviour. In this paper, we turn this approach upside down, and use the trajectories of probe molecules (BOD-IPY-labelled dodecanoic acid) to provide images based on the lateral heterogeneity of probe-surface interactions.…”
