Site‐specific and endothelial‐mediated dysfunction of the alveolar‐capillary barrier in response to lipopolysaccharides

Abstract: Infectious agents such as lipopolysaccharides (LPS) challenge the functional properties of the alveolar‐capillary barrier (ACB) in the lung. In this study, we analyse the site‐specific effects of LPS on the ACB and reveal the effects on the individual cell types and the ACB as a functional unit. Monocultures of H441 epithelial cells and co‐cultures of H441 with endothelial cells cultured on Transwells® were treated with LPS from the apical or basolateral compartment. Barrier properties were analysed by the tra… Show more

“…Furthermore, the gene expression of SP-B was decreased in cultured human pulmonary epithelial cells after treatment with conditioned medium from LPS stimulated macrophages, whereas in a monoculture of epithelial cells, LPS had not effect [77]. Janga et al [78] investigated the impact of LPS on monocultures of alveolar epithelial cells and microvascular endothelial cells, and a co-culture of both cell types, possibly imitating a functional alveolar-capillary barrier. Surprisingly, LPS had almost no effect on the barrier properties of epithelial cells in the monocultures.…”

“…e alveolar-capillary barrier is an important structure for maintaining gas exchange in the lung tissue. When this barrier is damaged, a large amount of fluid leaks and oxygen exchange in the lung tissue is impaired [17,18]. LPI and w/d ratio were used to observe the changes of lung permeability of rats in each group.…”

“…In addition, histopathological analysis showed that SARS-Cov-2 infection led to the induction of endothelitis in different organs. Finally, apoptosis and pyroptosis of endothelial cells were also highlighted [85].…”
