Site-specific monoubiquitination activates Ras by impeding GTPase-activating protein function

Abstract: SUMMARY Cell growth and differentiation are controlled by growth factor receptors coupled to the GTPase Ras. Oncogenic mutations disrupt GTPase activity leading to persistent Ras signaling and cancer progression. Recent evidence indicates that monoubiquitination of Ras leads to Ras activation. Mutation of the primary site of monoubiquitination impairs the ability of activated K–Ras to promote tumor growth. To determine the mechanism of human Ras activation we chemically ubiquitinated the protein and analyzed i… Show more

“…The catalytic domains of human SOS1 (SOS cat , residues 566 -1049) (26) and p120-RASGAP (GAP cat , residues 764 -981) were expressed in a pQlinkH vector (Addgene) and purified as described previously (33). cDNA sequences encoding the isolated RBDs of human BRAF (amino acids 149 -232) and CRAF (amino acids 51-132) were subcloned in a pET28a bacterial expression vector encoding an N-terminal 6-histidine tag and TEV cleavage sites and subsequently expressed in BL21 (DE3) cells.…”

“…While ubiquitination of H-Ras promotes endosomal localization, 25,26 monoubiquitination at Lys 147 in K-Ras upregulates K-Ras activity by impairing GAP-mediated GTP hydrolysis. 27,28 Moreover, a subset of Rho GTPases is ubiquitinated. Of these, polyubiquitination of RhoA, Rac1, and Cdc42 result in their deactivation and degradation.…”

“…Although no function has yet been assigned to monoubiquitination of Rac1, it is possible that this modification is involved in the mechanism by which Rho interacts with downstream effectors. Monoubiquitination of mammalian Ras proteins can lead to cell transformation (71) and does so by promoting nucleotide exchange or by impeding the binding of the GTPase activating protein (72,73).…”
