A marine bacterial strain, designated FR1311 T , was isolated from a sea-water sample from Ganghwa Island, South Korea. Cells were Gram-negative, facultatively anaerobic, catalase-and oxidase-positive, motile, oval or rod-shaped and halophilic (optimum sea-salt concentration for growth of 5-6 %). Phylogenetic analysis of its 16S rRNA gene sequence revealed that it represented a distinct line of descent within the genus Photobacterium. The major fatty acids were straight-chain saturated (C 16 : 0 ) and monounsaturated fatty acids (C 16 : 1 v7c and C 18 : 1 v7c). The predominant respiratory lipoquinone was Q-8. The DNA G+C content was 44 mol%. The phenotypic features of strain FR1199 T were similar to those of Photobacterium damselae subsp. damselae and Photobacterium damselae subsp. piscicida, but several physiological and chemotaxonomic properties readily distinguish the new isolate from them. On the basis of the polyphasic results revealed in this study, FR1311 T is considered to be the type strain of a novel species, for which the name Photobacterium ganghwense sp. nov. is proposed. The type strain is FR1311 T (=IMSNU 60287 T =KCTC 12328 T =JCM 12487 T ).

A species-specific plasmid (pZP1) has been isolated from the fish-pathogenic bacterium Pasteurella piscicida.Two DNA fragments of PZP1 (PZP1-1 with 964 bp and PZP1-4 with 477 bp) were cloned from the plasmid pZP1. Two 20-mer primer sets, PZP1-1a and PZP1-1b, and PZP1-4a and PZP1-4b, were constructed according to the nucleotide sequences of fragments PZP1-1 and PZP1-4.A 484-bp DNA fragment was amplified from template DNA from 40 strains of P. piscicida by PCR using the PZP1-1 a/1b primer set. The strains were isolated at different times of the year at different places in Japan and the USA. A 321-bp PCR product was amplified from all of the above strains of P. piscicida except strain ATCC17911 using the PZP1-4a/4b primer set. No such PCR products were obtained from template DNAs of Beneckea proteolytica, Photobacterium damsela, Ph. histaminum, Ph. leiognathi, 15 standard strains of Vibrio spp. or four fish pathogenic bacteria (Aeromonas hydrophila, A. salmonicida, Edwardsiella tarda and Enterococcus seriolicida).The PCR products were amplified with the PZP1-1a/1b primer set from DNA from the kidney of yellowtail (Seriola quinqueradiata) infected naturally with P. piscicida, but not from DNA from healthy yellowtail.

A multiplex-PCR approach, employing 2 primer pairs directed to internal regions of the 16s rRNA and ureC genes, was utilized to analyze a collection of Photobacterium damselae strains, including 25 isolates of subspecies piscicida and 15 isolates of subspecies darnselae. With this procedure, all the P. darnselae subsp. damselae strains yielded 2 amplification products, one of 267 bp and the other of 448 bp, corresponding to internal fragments of the 16s rRNA and ureCgenes, respectively. However, P. damselae subsp. piscicida isolates only showed the PCR product of 267 bp (16s rRNA fragment), indicating the absence of the urease gene in its genome. We have constructed a DNA probe directed to an internal region of the ureC gene, and corroborated by dot blot hybridization that the P. darnselae subsp. piscicida lacks this gene, whereas it is present in the subspecies darnselae. This constitutes the first successful discrimination between both subspecies using a PCR procedure, which could become a useful tool for diagnosis of pasteurellosis in the field. In addition, since these 2 subspecies have been shown to share nearly the same rrn operon sequence, our results provided evidence that one of the steps in the P, damselae speciation proccess included gainfloss events associated with the ure operon.
