Smart Injectable Chitosan Hydrogels Loaded with 5-Fluorouracil for the Treatment of Breast Cancer

Abstract: The treatment of breast cancer requires long chemotherapy management, which is accompanied by severe side effects. Localized delivery of anticancer drugs helps to increase the drug concentration at the site of action and overcome such a problem. In the present study, chitosan hydrogel was prepared for local delivery of 5-Fluorouracil. The in vitro release behavior was investigated and the anticancer activity was evaluated against MCF-7 cells using MTT assay. The in vivo studies were investigated via intra-tumo… Show more

“…Hydrogels based on CHT are gaining considerable interest for biomedical applications due to its biocompatibility, biodegradability, non-toxicity, and biological features, such as bio-adhesiveness, antibacterial, hemostatic, and anti-inflammatory properties. Altogether, these properties are pivotal in the design of biomedical systems for smart drug delivery [ 127 , 128 , 129 , 130 , 131 , 132 , 133 , 134 , 135 ], wound healing [ 136 ], and regenerative medicine [ 137 ].…”

“…In addition, a notable volume reduction in the TNBC spheroids is achieved, whereas free drugs only reduce growth rate [ 198 ]. Moreover, 5-Fluorouracil loaded chitosan hydrogel exhibits good physicochemical properties and in vitro sustained release profiles, resulting in greater inhibition of breast cancer cell viability and in vivo antitumor activity with minor side effects [ 199 ]. In addition, gefitinib-loaded cellulose acetate butyrate nanoparticles incorporated into chitosan/ÎČGP hydrogels is established for intratumoral administration in mice bearing breast cancer and displayed a greater cytotoxic effect in comparison with a free drug.…”

“…Because of the limitations of traditional chemotherapy and the toxicity of chemotherapeutic drugs to normal tissues and organs in oral cancer, the smart drug delivery system based on hydrogels has become a popular research direction. Related studies have mainly focused on the methods of sustained-release drug delivery and stimulation response [ 178 , 179 , 180 ]. The smart hydrogels have precise reactions to temperature, pH, and tiny changes in light.…”
