Smart pH- and Temperature-Sensitive Micelles Based on Chitosan Grafted with Fatty Acids to Increase the Efficiency and Selectivity of Doxorubicin and Its Adjuvant Regarding the Tumor Cells

Abstract: The main factors that determine the low effectiveness of chemotherapy are the low target bioavailability of antitumor drugs and the efflux process. In attempts to overcome this problem, several approaches are proposed here. Firstly, the development of polymeric micellar systems based on chitosan grafted by fatty acids (different types to optimize their properties), which, on the one hand, increase the solubility and bioavailability of cytostatics and, on the other hand, effectively interact with tumor cells du… Show more

“…Cytostatics 1, 2, 4 (Table 1, Scheme 1) were obtained according to the techniques described by the authors earlier with 1 H and 13 C NMR spectra [12,60]. The acid-modified chitosans Chit5-SA, Chit5-OA, Chit5-MUA and Chit5-LA were synthesized by the coupling reaction of COOH of acids with the NH 2 group using 1-ethyl-3-(3-dimethylaminopropyl) carbodiimide (EDC) and N-hydroxysuccinimide at 60 • C for 12 h (Figure S1), as described earlier [34].…”

“…Polymeric micelles based on chitosan modified with fatty acids are promising for strengthening anti-cancer and antibacterial drugs [17,18,23,24,27,29,30,[32][33][34][35][36][37][38][39]. In this article, a relatively new approach is proposed using the properties of chitosan as a polycation with pKa 6.4 [40,41], ionized in such a way that, in the microenvironment of tumors, the loaded drug is quickly released by the delivery system [34,[42][43][44][45][46]. In addition, chitosan can provide thermal sensitivity due to hydrophobic chain interactions at 38-42 • C. Fatty acids are necessary for the synthesis of amphiphilic polymers, forming polymer micelles which have low critical concentrations of micelle formation, are biocompatible and are highly selective.…”

“…In [ 26 ], it was shown that the peak of the fluorescence emission of rhodamine 6G when loaded into chitosan nanocomposite widens and shifts to the long-wavelength region. The changes in the fluorescence parameters when fluorophores molecules are loaded into chitosan-based micelles were previously demonstrated by us [ 35 , 36 ].…”

“…The synthesis of modified chitosan was carried out as described by us earlier with some modifications [ 45 , 46 , 47 ]. Chitosan was dissolved in 1 mM HCl solution (10 mg/mL) and then the pH was adjusted to 7.4 using 0.1 M phosphate buffer.…”

“…FTIR spectra of Chit5, LA, OA, Chit5-LA, and Chit5-OA were recorded using an FTIR microscope MICRAN-3 and Bruker Tensor 27 spectrometer equipped with a liquid-nitrogen-cooled MCT (mercury cadmium telluride) detector, as described earlier [ 45 , 48 ].…”
