SMYD Proteins: Key Regulators in Skeletal and Cardiac Muscle Development and Function

Abstract: Muscle fibers are composed of myofibrils, one of the most highly ordered macromolecular assemblies in cells. Recent studies demonstrate that members of the Smyd family play critical roles in myofibril assembly of skeletal and cardiac muscle during development. The Smyd family consists of five members including Smyd1, Smyd2, Smyd3, Smyd4, and Smyd5. They share two highly conserved structural and functional domains, namely the SET and MYND domains involved in lysine methylation and protein-protein interaction, r… Show more

“…SMYD2 regulates HSP90 methylation50 and the inhibition of HSP90 has been shown to reduce AAA formation in murine models51, suggesting this as a possible link between SMYD2 and AAA. SMYD2 also plays a role in the differentiation of embryonic stem cells52, again suggesting a possible role for aberrations of in utero aortic development influencing the risk of aortic disease later in life.…”

“…[12][13][14] The myogenic factor 6 (MYF6) gene belongs to the family of muscle regulatory factors (MRFs) and plays a major role in muscle growth and development. 15 MYF6 is the principal factor influencing the skeletal muscle phenotype.…”

“…Expression of two other muscle-enriched proteins, Smyd1, a member of a family of SET-MYND domain-containing lysine methyltransferases (10), which are critical for cardiac and skeletal muscle development (18), and Naca, whose musclespecific isoform, Sknac, is the major binding partner of Smyd1 in striated muscle (41), increased after removal of SB202190 from DM. Sknac knockout mice display impaired muscle growth and regeneration (41).…”
