SMYD3 contributes to a more aggressive phenotype of prostate cancer and targets Cyclin D2 through H4K20me3

Abstract: Prostate cancer (PCa) is one of the most incident cancers worldwide but clinical and pathological parameters have limited ability to discriminate between clinically significant and indolent PCa. Altered expression of histone methyltransferases and histone methylation patterns are involved in prostate carcinogenesis. SMYD3 transcript levels have prognostic value and discriminate among PCa with different clinical aggressiveness, so we decided to investigate its putative oncogenic role on PCa.We silenced SMYD3 an… Show more

“…Our results discovered that knockdown of NEAT1 expression remarkably decreased the protein expression of cyclin D1, while NEAT1 over-expression remarkably increased the level of cyclin D1 in A549 and H1299 cells (Figure 2G-2H). Cyclin D2 is highly expressed and promotes tumorigenesis in numerous of tumors [49, 50]. In our research, the protein expression of cyclin D2 was up-regulated by over-expression of NEAT1 (Figure 2G-2H).…”

“…Our results discovered that miR-134 significantly inhibited the protein expression of cyclin D1, while loss of miR-134 remarkably increased the level of cyclin D1 in A549 and SPC-A-1 cells (Figures 6H and 6I). Cyclin D2 is highly expressed and promotes tumorigenesis in numerous tumors [44, 45]. In our research, the protein expression of cyclin D2 was repressed by over-expression of miR-134 (Figures 6G and 6H).…”

“…Our results discovered miR-875-5p significantly inhibited the protein expression of EGFR, while loss of miR-875-5p remarkably increased the level of EGFR in HCT116 and SW480 cells (Figure 6K). cyclin D2 is highly expressed and promotes tumorigenesis in numerous tumors [30, 31]. In our research, the protein expression of cyclin D2 was repressed by over-expression of miR-875-5p (Figure 6K).…”
