SNP Discovery Using BSR-Seq Approach for Spot Blotch Resistance in Wheat (Triticum aestivum L.), an Essential Crop for Food Security

Abstract: The pathogenic fungus, Bipolaris sorokiniana, that causes spot blotch (SB) disease of wheat, is a major production constraint in the Eastern Gangetic Plains of South Asia and other warm, humid regions of the world. A recombinant inbred line population was developed and phenotyped at three SB-prone locations in India. The single nucleotide polymorphism (SNP) for SB resistance was identified using a bulked segregant RNA-Seq-based approach, referred to as “BSR-Seq.” Transcriptome sequencing of the resistant paren… Show more

“…Several genomic regions for resistance to spot blotch have been reported on different chromosomes in earlier studies [2,19,21,23,25,26,34,36,49,50,52,53,[64][65][66][67][68][69]. Only four major QTLs are designated as Sb1 on Chromosome 7D [19], Sb2 on Chromosome 5B [20], Sb3 on Chromosome 3B [21], and Sb4 on Chromosome 4BL [22] are well described.…”

“…(2012) developed a new mapping strategy combining bulk segregant analysis and RNA-Seq called ‘BSR-Seq’, where transcripts are sequenced from extreme bulks, being a potential technique for marker discovery in large polyploid genome like wheat. Using BSR-Seq, five SB-resistance associated transcripts were identified on 5B and 3B chromosomes and their potential role in SB resistance were inferred ( Saxesena et al., 2022 ).…”
