Sodium nitroprusside-induced protein phosphorylation in intact rat aorta is mimicked by 8-bromo cyclic GMP

Abstract: The effects of sodium nitroprusside, 8-bromo cyclic GMP, 8-bromoguanosine 5'-monophosphate, 8-bromo cyclic AMP, dibutyryl cyclic AMP, and isoproterenol on incorporation of 32P into proteins in intact rat thoracic aorta were studied. Aortas were incubated in [32P]orthophosphate in order to label endogenous adenosine triphosphate. Agents were then added for various times and the tissues were homogenized and fractionated (100,000 x g for 60 min) into soluble and particulate fractions. Soluble and particulate frac… Show more

“…The increased synthesis of cyclic GMP with endothelium-dependent vasodilators is also associated with cyclic GMP-dependent protein kinase activation and dephosphorylation of myosin light chain (32,36). Indeed, the profiles of altered protein phosphorylation with two-dimensional polyacrylamide gel electrophoresis of extracts from vascular smooth muscle after treatment with nitrovasodilators, endothelium-dependent vasodilators, and 8-bromo cyclic GMP are similar (34)(35)(36). While these effects with nitrovasodilators and 8-bromo cyclic GMP are endothelium-independent, the effects ofendotheliumdependent vasodilators require the endothelium.…”

“…The increase in cyclic GMP in tracheal and vascular smooth muscle is associated with the activation ofcyclic GMP-dependent protein kinase and the phosphorylation of numerous smooth muscle proteins (32)(33)(34)(35). One of the smooth muscle proteins in which phosphorylation is decreased with nitrovasodilators or cyclic GMP analogues is myosin light chain (3,35,36).…”

“…The pD2 values and the oxyhaemoglobin in the rat aorta (Kashiwabara et al, 1991). nitroprusside and 8-bromo cyclic GMP (Rapoport et al, 1982;Draznin et al, 1986 (Figure 3 and Table 1), but its maximum effect was slightly greater than that of KRN2391. Thus, the structure-activity relationships of pyridine derivatives as K channel openers indicates that the nitroxy substituent is very important for this activity.…”

“…It is known that sGC mediates cGMP production, which prompted an investigation into whether increased levels of cGMP mediate down-regulation of BDNF secretion. Indeed, basal BDNF secretion was reduced by 42 Ϯ 5% by treating cultured neurons directly with 1 mM 8Br-cGMP, a highly membrane-permeable form of cGMP (48) (Fig. 4B).…”
