Soil Microbial Community Response to Nitrogen Application on a Swamp Meadow in the Arid Region of Central Asia

Abstract: Although a large number of studies have reported the importance of microbial communities in terrestrial ecosystems and their response to nitrogen (N) application, it is not clear in arid alpine wetlands, and the mechanisms involved need to be clarified. Therefore, the response of the soil microbial community in a swamp meadow to short-term (1 year) N application (CK: 0, N1: 8, N2: 16 kg⋅N⋅hm–2⋅a–1) was studied using 16S/ITS rRNA gene high-throughput sequencing technology. Results showed that N application had … Show more

“…Soil microorganisms are sensitive to environmental changes, and their composition and activity are influenced by a variety of factors including fertilizations, climate, and plant type ( Dong et al, 2014 ; Soman et al, 2017 ; Grosso et al, 2018 ; Hu et al, 2019 ). Nitrogen fertilizer is an important factor that affects soil microbial communities in many agricultural systems ( Wang L. et al, 2021 ; Zhang X. et al, 2021 ; Hu et al, 2022 ). In this study, the bacterial community diversity and richness of the apple rhizosphere soil in the nitrogen application treatment were higher than those without nitrogen treatment.…”

“…A pH meter (Mettler Toledo FE28-Standard, Switzerland) was used to measure soil pH at water: soil ratio of 2.5:1. The H 2 SO 4 –K 2 Cr 2 O 7 external heating method ( Hu et al, 2022 ), alkali diffusion method, spectrophotometry (Shimadzu UV-1780, Japan; Shao et al, 2019 ), flame photometry (Shanghaiyuefeng FP6400, China; Chen et al, 2021 ), the drying method, H 2 SO 4 -HClO 4 digestion—spectrophotometry (Shimadzu UV-1780, Japan), H 2 SO 4 -HClO 4 digestion—flame photometry (Shanghaiyuefeng FP6400, China; Mei et al, 2021 ), and an elemental analyzer (Euro EA3000, Italy) were used to measure the content of SOC, AN, AP, AK, soil moisture content, total P (TP), total K (TK), and total N (TN), respectively.…”

“…At present, high-throughput sequencing technology has been widely utilized for the analysis of microbial community of diverse environmental samples (Dreier et al, 2022 ). Compared with traditional fingerprint technology such as restriction fragment length polymorphism (RFLP), amplified fragment length polymorphism (AFLP), and random amplified polymorphic DNA (RAPD), high-throughput sequencing by Illumina MiSeq sequencing platform has many advantages such as high-throughput, fast rate, and accurate result (Hu et al, 2022 ). High-throughput sequencing has also been widely used to analyze the microbial community of various Daqu samples (Deng et al, 2020 ; Chen et al, 2021 ).…”
