Solid Lipid Nanoparticles: An Approach to Improve Oral Drug Delivery

Abstract: Nanoparticles have shown overall beneficial effects in drug administration. Specifically, solid lipid nanoparticles (SLN) have emerged as an alternative to polymer-based systems. However, the oral administration of SLN, the first choice for conventional medications, has not been addressed due to the taboo surrounding the complicated transit that this delivery route entails. This review focuses on the encapsulation of drugs into SLN as a strategy for improving oral administration. Examples of applications of SL… Show more

“…SLNs have been widely applied for delivery of drugs for neurological diseases and cancer, and for preventing antibiotic resistance, because this carrier can contain many antimicrobial drugs, increase drug absorption, and reduce bacterial efflux. SLNs are a good alternative in terms of functionality and biocompatibility, because they can directly reach the site of infection, increase drug bioavailability, and reduce toxicity or side effects [ 122 ].…”

“… SLNs are being broadly studied for delivering anti-cancer drugs by incorporating them into an SLN formulation to improve oral bioavailability of drugs, protecting them from labile environment & also helpful in reducing adverse effects by minimizing the dosage by effective targeting to site of action 27 . There is various drugs approved by FDA  Additional medical study on lipid nanoparticles for releasing mRNA to solid cancer cells or lymphoma is presently underway (NCT03739931).…”

“…SLNPs [ 56 , 57 ], similarly to polymer-based nanoscale delivery systems, can improve the absorption and bioavailability of drugs with a low solubility and can protect molecules, which are labile in an acidic environment and can enable the targeting of drugs to their side of action and can reduce adverse side effects [ 169 ]; the emergence of antibiotic resistance can be reduced by the incorporation of antibiotics into SLNPs [ 170 ]. Ascorbyl tocopherol succinate, which is used as an adjuvant in SLNPs loaded with VAN, showing a mean size of 106.9 ± 1.4 nm and a zeta potential of −16.5 ± 0.93 mV, contributed to a pronounced increase in drug release in an acidified environment compared to controls; as a response to the lipase enzyme, these antibiotic-loaded SLNPs caused double higher growth inhibition of MRSA biofilm for 5 days and a 3.44-fold reduction of bacteria in a skin-infected mice model compared to free drug [ 171 ].…”
