Somatic Embryogenesis in Two Orchid Genera (Cymbidium, Dendrobium)

Abstract: The protocorm-like body (PLB) is the de facto somatic embryo in orchids. Here we describe detailed protocols for two orchid genera (hybrid Cymbidium Twilight Moon 'Day Light' and Dendrobium 'Jayakarta', D. 'Gradita 31', and D. 'Zahra FR 62') for generating PLBs. These protocols will most likely have to be tweaked for different cultivars as the response of orchids in vitro tends to be dependent on genotype. In addition to primary somatic embryogenesis, secondary (or repetitive) somatic embryogenesis is also des… Show more

“…In this medium, periodic subcultures (4-5 times, each subculture period was 15 days) were carried out in the same medium to proliferate PLBs. For more details, also see Teixeira da Silva et al (2015a) and Teixeira da Silva and Winarto (2016). Fourth, bacterial (I-O) and fungal (P-U) contamination of Dendrobium 'Zahra FR 62' (I, J, L, M, O, R), D. 'Gradita 31' (K, N, P, S) and D. 'Jayakarta' (Q, T) in solid and semi-solid cultures of shoot tips (I-K) or PLBs (L-T) can take place 2 months (I-Q) or 1.5 months (R-T) after culture initiation on half-strength MS containing 0.3 mg dm -3 TDZ and 0.1 mg dm -3 NAA.…”

“…Besides the influence of light, another admittedly important factor in the induction of PLBs in Phalaenopsis and orchids in general is the genotype [61]. This means that under the same cultivation condition, the induction responses of PLBs may be significantly different [62], which is still considered a limitation of the technique.…”

“…Regarding the type of explant, leaf segments of plants grown in vitro have been the most suitable for induction of PLBs in Phalaenopsis (45% of papers; Table 1), but there are reports of protocols that Besides the influence of light, another admittedly important factor in the induction of PLBs in Phalaenopsis and orchids in general is the genotype [61]. “…There are two pathways in somatic embryogenesis. First, single-cellderived somatic embryo is generated, develops into a globular somatic embryo similar to zygotic embryogenesis (27) which later become PLBs with shoot tip, and eventually develops into plantlets (10).…”
