In recent years, the application of natural substances on crops has been intensified in order to increase the resistance and yield of the soybean crop. Among these products are included plant biostimulants that may contain algae extracts, amino acids, and plant regulators in their composition. However, there is little information on the isolated effect of each of these constituents. The objective of this research was to evaluate the effect of the application of isolated amino acids on the antioxidant metabolism of the soybean crop. Experiments were carried out in a greenhouse and in the field with the application of the amino acids glutamate, phenylalanine, cysteine, glycine in seed treatment, and foliar application at V4 growth stage. Antioxidant metabolism constituents evaluated were superoxide dismutase, catalase, peroxidase, hydrogen peroxide content, proline, and lipid peroxidation. In addition, resistance enzymes as polyphenol oxidase and phenylalanine ammonia-lyase (PAL) were evaluated. In both experiments, the use of cysteine, only in seed treatment and in both seed treatment and foliar application increased the activity of the enzyme PAL and catalase. Also in both experiments, the use of phenylalanine increased the activity of the enzyme PAL when the application was carried out as foliar application or both in seed treatment and foliar application. In the field experiment, the application of glutamate led to an increase in the activity of the catalase and PAL enzymes for seed treatment and foliar application. The use of the set of amino acids was only efficient in foliar application, which led to a greater activity of the enzymes peroxidase, PAL, and polyphenol oxidase. The other enzymes as well as lipid peroxidation and hydrogen peroxide presented different results according to the experiment. Therefore, glutamate, cysteine, phenylalanine, and glycine can act as signaling amino acids in soybean plants, since small doses are enough to increase the activity of the antioxidant enzymes.

Plant SOS1 gene encodes plasma membrane Na + /H + antiporter, which helps in the exclusion of Na + and improves plant salt tolerance. However, detailed studies of SOS1 gene in the important oil crop, soybean (Glycine max), are still lacking. In the present study, we carried out a comprehensive bioinformatics analysis of SOS1 gene in soybean.Referring to the analysis of physicochemical properties and structural characteristics, the GmSOS1 is an acidic protein with instability and hydrophobicity. Subcellular localization of GmSOS1 supports our presumption that GmSOS1 protein is a plasma membrane Na + /H + antiporter. The prediction of phosphorylation sites indicates 4 sites that may be activated. Further, the functional protein association network and co-functional network signify the potential role of GmSOS1 in salt stress tolerance. Although the interaction between GmSOS1 and GmHKT1 remains elusive, some of the intermediary signaling components of SOS pathway in soybean have been predicted. In addition, in silico expression analysis based on transcriptome datasets using publicly available database revealed that GmSOS1 was differentially expressed in tissues and different times. Due to the analysis of its regulation mechanism, we found transcription factors such as WRKY and ERF as well as three miRNAs can regulate the expression of GmSOS1. Phylogenetic analysis using the homologous amino acid sequence of SOS1s from 26 species was performed to study the conserved motifs among these SOS1 members. Overall, we provide an extensive analysis of the GmSOS1 gene and it promises the primary basis for the study in development and response to salt tolerance.

Glycine max cultivars Lee68, Nannong1138-2, Nannong8831 were used as the female parents, and hybrid lines (F 5) 4111, 4076 [N23674 × BB52], 3060 [Lee68 × N23227] and 185 [Jackson × BB52] that selected for salt tolerance generation by generation from the cross combination of G. max and G. soja were used as the male parents, 11 (A-K) backcrosses or three-way crosses were designed and 213 single hybrids were harvested. The optimized soybean SSR-PCR system was used to analyze the SSR polymorphism of above parental lines and get the parental co-dominant SSR markers for hybrid identification, and in which 30 true hybrids were gained. The true hybrids (G1, G3, G9, G12, G13, G16) of G cross combination were chosen as the representative for the salt tolerance test, and the results showed that, as exposed to salt stress, the seedlings of G9 line displayed higher salt tolerant coefficient, relative growth rate and dry matter accumulation, when compared with their female parent Nannong1138-2, and even performed equally strong salt tolerance as the male parent 3060. It provides a feasible method of the combination of molecular SSR markers and simple physiological parameters to identify the true hybrids of G. max and G. soja, and to innovate the salt-tolerant soybean germplasms.
