Spatial determination of elements in green leaves of oak trees (Quercus robur) by laser ablation-ICP-MS

Abstract: Laser ablation inductively coupled plasma mass spectrometry (laser ablation-ICP-MS) has been applied to the spatially resolved determination of the elements Mg, Ca, Cu, Ni, Ba, Al, Pb, Sr and Mn in green leaves of oak trees. Instrument operating parameters such as the laser wavelength and the pulse energy have been optimized to provide the sensitivity and reproducibility required for the analysis. The method provides spatial resolution down to 300 microm with the use of the 355 nm wavelength (3rd harmonic of t… Show more

“…The spectroscopic laser ablation method has been used to find spatial patterns in cation concentrations in green oak leaves (Hoffmann et al 2000). Our results confirm that good sensitivity and fine spatial resolution, combined with the ability to sample growing tissue with minimal disruption to surrounding tissue (Fig.…”

“…www.rsc.org/jaas for quantification purposes in LA-ICP-MS using a preparation of matrix-matched laboratory standards or by solution-based calibration. Due to these advantages, LA-ICP-MS, which is mostly used as a trace analytical technique for the bulk analysis of solid inorganic matrices and partly for isotope ratio measurements, [16][17][18][19] will also be applied to an increasing extent for element distribution analysis (including analysis of trace elements) in biological samples and medical tissues (e.g., in tree barks, 20 green leaves, 21 fish 22 or liver tissue 13 ). In addition, LA-ICP-MS was applied as a microlocal analytical method for the determination of P, S, Si and metal concentrations (Al, Zn, Cu und Fe) in well-separated protein spots after two-dimensional gel electrophoresis in brain samples of patients with Alzheimer's disease and in control brains.…”

“…However, when a LA system is coupled to an inductively coupled plasma mass spectrometer (ICP-MS), the ablated material is fully ionized in the argon plasma and the elements are detected by an element-specific detector (12 ). Only a few studies, however, have used this method for soft tissues (13,14 ). In our previous study we used LA coupled to inductively coupled plasma time-of-flight mass spectrometry (ICP-TOF-MS) for the quantification of trace elements in soft tissues.…”
