Spatial distribution of CD115+ and CD11b+ cells and their temporal activation during oxygen-induced retinopathy in mice

Abstract: Using two different approaches to monitor MP/MG cells, our findings demonstrated that MP/MG concentrate within pathologically vascularized areas during OIR. We were able to clarify that reactive changes of CD11b cell distribution to OIR primarily occur in the deep retinal layers. Furthermore, we found the highest proportion of activated CD11b cells in regions with pathologic neovascularization processes. Our findings support previous reports about activated MP/MG guiding revascularization in avascular areas an… Show more

“…It is well known that the neovascular response in the OIR model is accompanied by upregulation of inflammatory signals, infiltration of mononuclear phagocytes and activation of resident microglia ( Binet et al, 2020 ; Brockmann et al, 2018 ; Davies et al, 2006 ; Sun et al, 2017 ; Wang et al, 2020 ). This relates to preterm human infants where the risk of ROP is associated with sepsis ( Huang et al, 2019b ) and systemic inflammation ( Lee and Dammann, 2012 ; Sood et al, 2010 ).…”

“…Statistical analysis was performed by one- way ANOVA (b, d, e) and the two-sided Mann-Whitney test (f, h), *p < 0.05 ◂ significantly reduced number of CD11b+ cells. This may result from a milder phenotype of retinal neovascularization in the early phases of the hypoxic phase due to initial improved revascularization in Vhl MCΔ/Δ retinas, with reduced drive to recruit CD11b+ cells to neovascular sites in OIR [27]. Although not statistically significant, our finding that Vhl MCΔ/Δ retinas tended to have reduced lectin + myeloid cells density in neovascular areas (Fig.…”
