Specific and sensitive plate assay for bacterial lipases

Abstract: A plate assay to detect bacterial lipase (EC 3.1.1.3) in a medium containing trioleoylglycerol and the fluorescent dye rhodamine B is presented. Substrate hydrolysis causes the formation of orange fluorescent halos around bacterial colonies visible upon UV irradiation. The logarithm of lipase activity from cell-free culture supernatants is linearly correlated with the diameter of halos, thereby allowing quantitation of lipase activities ranging from 1 to 30 nkat.

“…To overcome these limitations, Kouker and Jaeger [21] employed the fluorescence dye Rhodamine B to observe the zone of lipolysis as an orange fluorescence under UV light at 350 nm. Rhodamine forms a fluorescent complex with free fatty acids.…”

“…Presence of lipolytic activity in B. megaterium 370 was analysed using Rhodamine B-supplemented plates [11], containing tributyrin or olive oil. The strain showed good performance on lipid-based substrate hydrolysis, as con¢rmed by £uorescence emission from Rhodamine B and appearance of hydrolysis areas around the colonies (not shown).…”

“…Standard methods were used for Gram staining and testing for aminopeptidase, oxidation fermentation and gas formation, activities of oxidase and catalase and gelatin hydrolysis [20]. Secretion of lipase was tested on rhodamine B medium [21]. Partial 16S RNA gene sequencing and sequence alignments were performed by DSMZ [22,23].…”
