Specific and straightforward molecular investigation of β-thalassemia mutations in the Malaysian Malays and Chinese using direct TaqMan genotyping assays

Abstract: ABSTRACT. Beta-thalassemia is a life-threatening inherited blood disorder. Rapid characterization of β-globin gene mutations is necessary because of the high frequency of Malaysian β-thalassemia carriers. A combination real-time polymerase chain reaction genotyping assay using TaqMan probes was developed to confirm β-globin gene mutations. In this study, primers and probes were designed to specifically identify 8 common β-thalassemia mutations in the Malaysian Malay and Chinese ethnic groups using the Primer E… Show more

“…In addition to Hb E, the β-globin mutations reported in this study, i.e. CD 17 (A-T), CD 41/42 (-TTCT), and nucleotide -28 (A-G), were also similar to those reported in neighboring countries [5,[19][20][21] . Interaction of these abnormal genes can lead to several complex thalassemia syndromes, reflecting the serious health burden within the country.…”

“…In this context, the TaqMan ® genotyping assay represents a reliable, sensitive and cost-effective diagnostic method allowing the discrimination of single nucleotide polymorphisms [ 34 , 35 ]. Thus, the TaqMan ® genotyping assay is suitable for large-scale screenings and high-throughput analyses.…”

“…TaqMan ® genotyping assays were designed for characterization of β-thalassemia mutations in the Malays and Kingdom of Saudi Arabia [ 14 , 34 36 ]. Also, an analogous TaqMan ® assay was developed and reported to be effective for detection of the Sardinian Wilson’s mutation [ 37 ].…”

“…In the area of result interpretation, genotyping using Bio‐Rad CFX manager shows quadrant separation in the allelic discrimination plots that are different from the scatter plots in the studies carried out by Kho et al . . Quadrant separation genotypes the samples into four different columns that consist of wild‐type, heterozygous, homozygous and DNA blank.…”
