Specific Magnetic Isolation of E6 HPV16 Modified Magnetizable Particles Coupled with PCR and Electrochemical Detection

Abstract: The majority of carcinomas that were developed due to the infection with human papillomavirus (HPV) are caused by high-risk HPV types, HPV16 and HPV18. These HPV types contain the E6 and E7 oncogenes, so the fast detection of these oncogenes is an important point to avoid the development of cancer. Many different HPV tests are available to detect the presence of HPV in biological samples. The aim of this study was to design a fast and low cost method for HPV identification employing magnetic isolation, polymer… Show more

“…According to the manufacturer’s instruction (Thermo Fisher Scientific, Waltham, MA, USA), the biotinylated DNA probe complementary to LINC00657 was incubated with Dynabeads M-280 Streptavidin after being dissolved in binding and washing buffer for 10 min at room temperature [ 13 ]. After incubation, the probe-coated beads were generated.…”

“…Jimanez et al in 2016 developed a platform sensor based on magnetizable particles (MPs) for the diagnosis of purified HPV16‐E6 gene from PUK57 plasmid by SWV technique. Actually, MPs modified by primers coupled with electrochemical and electrophoretic gel detection of the isolated nucleic acid in combination with PCR method acted in a concentration range from 0.15 to 2.5 μM and the LOD of 0.2 nM .…”

“…Another method, based on magnetizable nanoparticles and electrochemical detection has been designed for the detection of E6-HPV16 gene, which has been demonstrated to be implicated in mechanisms of human papilloma virus (HPV)-related carcinogenesis. Its combination with PCR improved DNA isolation in comparison with standard PCR, allowing detection of a lower oncogene level [ 90 ].…”
