Spontaneous atopic dermatitis in mice expressing an inducible thymic stromal lymphopoietin transgene specifically in the skin

Abstract: The cytokine thymic stromal lymphopoietin (TSLP) has recently been implicated in the pathogenesis of atopic dermatitis (AD) and other allergic diseases in humans. To further characterize its role in this disease process, transgenic mice were generated that express a keratinocyte-specific, tetracycline-inducible TSLP transgene. Skin-specific overexpression of TSLP resulted in an AD-like phenotype, with the development of eczematous lesions containing inflammatory dermal cellular infiltrates, a dramatic increase… Show more

“…The mice were challenged intranasally with PBS in a similar manner for the negative control. During the OVA challenge, rat antimouse TSLP neutralizing antibody or the isotype control rat IgG2a are consistent with the previous in vivo studies using mouse models of asthma and atopic dermatitis [5][6][7]. Thus, the current results also support the importance of TSLP in allergic inflammation.…”

“…For instance, transgenic mice expressing TSLP in keratinocytes or in lung epithelial cells develop atopic dermatitisor asthma-like inflammation in the skin or the lung, respectively, while TSLPR-deficient mice fail to develop an inflammatory lung response to inhaled antigen [5][6][7].…”

“…The well known cytokine thymic stromal lymphopoietin (TSLP) produced by keratinocytes activates DCs which in turn primes T cells to support the maintenance of Th2 cells. 84 Chemokines like RANTES are produced in significant amounts following stimulation with TNF-α and IFN-γ leading to increased recruitment of eosinophils. 85,86 Epidermal keratinocytes of AD lesions also produce increased levels of GM-CSF.…”
