SPP1 promotes Schwann cell proliferation and survival through PKCα by binding with CD44 and αvβ3 after peripheral nerve injury

Abstract: Background: Schwann cells (SCs) play a crucial role in Wallerian degeneration after peripheral nerve injury. The expression of genes in SCs undergo a series of changes, which greatly affect the proliferation and apoptosis of SCs as well as the fate of peripheral nerve regeneration. However, how do these genes regulate the proliferation and apoptosis of SCs remains unclear. Results: SPP1 and PKCα were found upregulated after human median peripheral nerve injury, which promoted SCs proliferation and survival. Th… Show more

“…Interestingly, SPP1 is known for its anti-apoptotic properties in numerous cell types. 39 , 40 One could speculate that anti-apoptotic macrophage signalling via SPP1 would lead to paracrine, pro-fibrotic signalling. Interestingly, pro-apoptotic BNIP3 mRNA was enhanced across all macrophage subsets, while SPP1 up-regulation was most pronounced in TREM2hi macrophages.…”

“…Osteopontin upregulated after peripheral nerve injury has been showed to promote proliferation and inhibit apoptosis of Schwann cells. 70 Increased myelin basic protein synthesis and enhanced myelin formation were also observed in vitro after the Spp1 treatment. 71 The interaction between aged microglia and OLG C2 (such as Spp1 ~ Itgb5, Spp1 ~ Itgav ) may provide a therapeutic target for promoting remyelination in aged.…”

“…As a result, we obtained 66 receptor-ligand pairs that may play a role in the five main differentiation trajectories mentioned above ( Figures S4 A and S4B). The SPP1_CD44 interaction showed high intensity in the 11th week and 17th week of gestation and was found in all three neuroectodermal progenitor differentiation processes, suggesting its role as a typical marker of neuroectodermal progenitor differentiation ( Wang et al., 2020 ) ( Figures 5 E, S4 D, and S4G). Both EPHA3_EFNA4 and LGR4_RSPO3 interactions, which were related to the formation of muscle stem cells in mice ( Alonso-Martin et al., 2016 ) and differentiation of hASCs ( Zhang et al., 2017a ), respectively, were only observed between immune cells and fibroblast subclusters ( Figure 5 E).…”
