Star‐Shaped Polypeptides: Synthesis and Opportunities for Delivery of Therapeutics

Abstract: Significant advances in the synthesis of polypeptides by N-carboxyanhydride (NCA) polymerisation over the last decade have enabled the design of advanced polypeptide architectures such as star-shaped polypeptides. These materials combine the functionality offered by amino acids with the flexibility of creating stable nanoparticles with adjustable cargo space for therapeutic delivery. This review highlights recent advances in the synthesis of star polypeptides by NCA polymerisation followed by a critical review… Show more

“…In addition, the microstructure of the side chains can be controlled to yield core–shell structures with compartments of different polarity. This precise control over size and internal structure may provide solutions to tackle some of the limitations of other drug delivery systems, for example in cancer therapy, where intratumoral pressure substantially limits their penetration . In general, a nanodrug delivery system (NanoDDS) for chemotherapeutics designed with regard to cancer therapy needs to deliver its cargo to the tumor cells.…”

“…Synthetic polypeptides derived from the polymerization of amino acid N ‐carboxyanhydrides (NCA) are promising biomimetic materials with potential for biomedical applications . Advanced polymerization techniques in tandem with recently developed orthogonal functionalization approaches, have enabled a large structural and functional diversity with the potential to design bespoke polypeptide biomaterials .…”

“…It was envisaged that due to its poly(cationic) nature, poly[(PheLA) 10.4 ‐ b ‐(LysLA) 31.4 ] may be exploited for antimicrobial applications and gene delivery . However, its polycationic nature also dictates that this PHA could pose cytotoxicity problems to healthy cells if it were to be used for drug delivery in vivo .…”
