STAT3-Inducible Mouse ESCs: A Model to Study the Role of STAT3 in ESC Maintenance and Lineage Differentiation

Abstract: Studies have demonstrated that STAT3 is essential in maintaining self-renewal of embryonic stem cells (ESCs) and modulates ESC differentiation. However, there is still lack of direct evidence on STAT3 functions in ESCs and embryogenesis because constitutive STAT3 knockout (KO) mouse is embryonic lethal at E6.5–E7.5, prior to potential functional role in early development can be assessed. Therefore, in this study, two inducible STAT3 ESC lines were established, including the STAT3 knockout (InSTAT3 KO) and pSTA… Show more

“…A remaining question is the molecular mechanisms through which TNFR2 mediates CSC proliferation, differentiation and maturation. Recent studies suggest that both Akt and STAT3 are critical for CSC proliferation and differentiation from ESCs. We observe that both Akt and STAT3 are highly activated at early phase of CSC differentiation.…”

“…The important role of STAT3 in cardiac differentiation is also supported by a newly published study. It was discovered that deletion of STAT3 in ESCs would affect the differentiation of ESCs into the mesoderm and cardiac lineage [57]. The mechanisms through which STAT3 regulates CPC generation and cardiac differentiation need to be elucidated in detail in the future work.…”

“…STAT3, a member of signal transducers and activators of transcription (STAT) family of transcription factors, regulates various cellular function, including embryogenesis, inflammation, and immunity [28,29]. Despite its crucial role in the physiological processes, STAT3 also promotes tumor development and progression.…”
