STAT4 is required for the generation of Th1 and Th2, but not Th17 immune responses during monophosphoryl lipid A adjuvant activity

Abstract: STAT4 is critical for the production of IFN-γ during the generation of T1 immune responses. We investigated the role of STAT4 in mediating T1-inducing activity of a vaccine adjuvant monophosphoryl lipid A (MPL-A) using the standard antigen ovalbumin (OVA) in STAT4KO mice. Our results show that splenocytes from STAT4KO mice displayed lower OVA-specific T-cell proliferation and IL-2 production compared with wild-type (WT) mice. Further, IFN-γ production was diminished in STAT4KO-derived splenocytes but the level… Show more

“…STAT4 signaling is induced by a number of cytokines including IL-12 and IL-2 [ 161 ] and is critical for IFNγ production during generation of Th1 responses [ 162 ]. STAT3 signaling is induced by a number of cytokines including IL-6 and OSM (and growth factors such as GM-CSF), with BCG vaccination recently shown to cause STAT3 phosphorylation in antigen presenting cells [ 163 ].…”

“…Moreover, it was recently discovered that the silencing of STAT4 led to a reduction in the serum levels of IFN-γ and IL-2, with an elevation in the serum levels of IL-6 and IL-10 ( 43 ). IFN-γ production was lowered in STAT4 KO-derived splenocytes, but no significant differences in the levels of IL-12 and TNF-α were found when compared to those in WT mice ( 44 ). The effect of STAT4 reduction on VDR expression should be further elucidated in future studies.…”
