Stem cell therapy for Alzheimer’s disease: An overview of experimental models and reality

Abstract: Alzheimer's disease (AD) is a neurodegenerative disorder. The pathology of AD is characterized by extracellular amyloid beta (Aβ) plaques, neurofibrillary tangles composed of hyperphosphorylated tau, neuronal death, synapse loss, and brain atrophy. Many therapies have been tested to improve or at least effectively modify the course of AD. Meaningful data indicate that the transplantation of stem cells can alleviate neuropathology and significantly ameliorate cognitive deficits in animal models with Alzheimer's… Show more

“…BM-derived immature monocytic cells can commit to a microglia-like phenotype, and they harbor several features and functions of native microglia [ 94 ]. Since resident microglia appear to degenerate in AD, Mr BMT compensates for the deficient functions of senescent resident microglia in AD [ 95 ]. ALS is a progressive, fatal neurodegenerative disease.…”

“…We speculated that novel Acr-based controllable tools for a wide range of applications can be developed using the PICI system with broad-spectrum inhibitors such as AcrIIC1 and AcrIIA5 ( 26 , 27 ). In particular, developing more advanced Acr-based tools to achieve precise, allele-specific and controllable genome editing may provide new opportunities for biomedical applications in the future ( 49 , 50 ).…”

“…Various types of stem cells have been used in stem cell therapy for AD, and implant procedures for stem cells have been developed [122]. Potential risks of oncogenesis, safety of long-term use, efficiency of reprogramming, and avoidance of immune rejection are considerable safety considerations for stem cell implantation, including that of iPS cells.…”
