Stemness Related Genes Revealed by Network Analysis Associated With Tumor Immune Microenvironment and the Clinical Outcome in Lung Adenocarcinoma

Abstract: Zeng et al. Stemness Genes Affect Immune Infiltration and immunity. The expression of SRGs could probably affect our tumor occurrence, progression, the efficacy of chemotherapy and immunotherapy, and clinical outcomes. In conclusion, the 15 SRGs reported in our study may be used as potential candidate biomarkers for prognostic indicators and therapeutic targets after further validation.

“…Nowadays, widespread analysis of high-throughput technologies via advanced computational techniques facilitates the identification of disease-related biomarkers ( Fernandes et al, 2020 ). Weighted gene co-expression network analysis (WGCNA) is a well-documented systematic bioinformatics tool to extract co-related gene modules and then identify hub genes that correlated with clinical traits of interest ( Horvath and Dong, 2008 ; Kakati et al, 2019 ), and has been widely applied for potential therapeutic target detections in various cancers ( Zeng et al, 2020 ; Zhang et al, 2020 ; Zhu and Hou, 2020 ). In this study, based on RNA-seq data acquired from public database, M2 macrophage fractions in stage III/IV pMMR CRC patients were calculated by the CIBERSORT algorithm, and WGCNA was then performed to identify the associated hub genes.…”

“…[ 51 ] Previous studies also studied the relationship between cancer stemness and anti‐tumor immunity and revealed that cancer stemness was negatively correlated with immune infiltration in LUAD. [ 52,53 ]…”

“…One of the explanations as to why pediatric tumors are less accessible for elimination by the immune system is stemness itself. The mRNAsi has been linked with immune cell exclusion and protection of antigenic clones from elimination [ 3 , 4 , 7 ]. Embryonic, mesenchymal and induced pluripotent stem cells possess immune modulatory properties, while resistance to immune-mediated destruction is an intrinsic feature of quiescent adult tissue stem cells [ 8 ] and cancer stem cells [ 9 , 10 , 11 ].…”
