Stimuli‐responsive nanotherapeutics for precision drug delivery and cancer therapy

Abstract: Cancer remains one of the world's leading causes of death. However, most conventional chemotherapeutic drugs only show a narrow therapeutic window in patients because of their inability to discriminate cancer cells from healthy cells. Nanoparticle-based therapeutics (termed nanotherapeutics) have emerged as potential solutions to mitigate many obstacles posed by these free drugs. Deep insights into knowledge of the tumor microenvironment and materials science make it possible to construct nanotherapeutics that… Show more

“…These benefits include improved epithelium permeability, enhanced stability, extended halflife, increased solubility and bioavailability, improved tissue targeting as well as minimized side effects (Date et al, 2016;Kermanizadeh et al, 2018). The nanotechnology-based systems are increasingly applied now to prevent and cure aging-associated pathological conditions, including neurodegenerative disorders such as Alzheimer's and Parkinson's diseases (Poovaiah et al, 2018;Ramanathan et al, 2018), cardiovascular diseases (Li et al, 2018;Taneja et al, 2019), obesity and type 2 diabetes (Bahadori et al, 2019;Tsou et al, 2019), and also tumors (Qiao et al, 2019).…”

“…When the rate of ROS production in the cells reached an undesired state, antioxidative machinery is activated to keep the cells in a proactive position. Therefore, having high capability for both oxidative and antioxidative machinery in cancer cells and sophisticated mechanisms over maintaining a balance between the two phenomenon has made cancer cells easily adopt to new environments for promotion of tumor growth (Qiao et al, 2018;Tsang et al, 2018). The role for melatonin here is to reduce the load of ROS on normal cells, instead, pushing this load toward cancer cells.…”

“…Studies have taken advantage of the differences between the tumor microenvironment and the normal environment to figure out ways to release drugs from these nanoparticles. is has mainly been via dissociation of chemical bonds or structures that are sensitive to the tumor microenvironment [124,125].…”
