Storage Characteristics of Split Double-Dose Platelet Concentrates Derived from Apheresis and Treated with Amotosalen Hydrochloride and UVA Light for Pathogen Inactivation

Abstract: Background: By inhibiting nucleic acids, photochemical treatment (PCT) using amotosalen hydrochloride (S-59) in combination with UVA light provides a new method for the inactivation of pathogens and leukocytes, potential contaminants of platelet concentrates (PCs). Material and Methods: We evaluated the effect of PCT on function and viability of PCs derived from apheresis over a storage period of 5 days. Double-dose PCs containing 6.5–7.0 × 1011 platelets (n = 8) were collected in 600 ml of approx… Show more

“…I on Day 8). Our data are in line with previous reports showing a storage‐related increase of P‐selectin expression 21‐24 and a decline in the PLTs' residual functional capacity to respond to agonists like TRAP‐6, 1,21 particularly after PRT treatment 14,17,25 . The data suggest also that storage and PRT treatment induces continuous depletion of P‐selectin from the alpha granules until exhaustion of the stores.…”

“…PRT treatment with the INTERCEPT Blood System (I units) and the MIRASOL‐PRT system (M units) was performed 2 hours after the end of collection in parallel according to the manufacturer's instructions as described previously 13‐18 . In brief, for photochemical treatment with the INTERCEPT Blood System, a commercially available device (Code R5628B, Cerus Corp.) consisting of a series of four closed interconnected plastic containers was used.…”

“…As a consequence, amotosalen phototreatment does not affect most PLT physiologic functions. This has been supported by in vitro 16‐18 and in vivo studies 19‐21 . Yet, PLTs contain viable mitochondria 22 and mitochondrial DNA (mtDNA).…”
