STR polymorphisms in Sri Lanka: evaluation of forensic utility in identification of individuals and parentage testing

Abstract: Objectives This preliminary study was carried out to determine the allele frequencies and forensic efficiency parameters for the short tandem repeat loci CSF1PO, TPOX, THO1, D16S539, D7S820, D13S317, vWA, FESFPS and F13B in a test sample population of Sri Lankans. Design Test samples were obtained from 305 nonrelated individuals originating from all 9 provinces of Sri Lanka. DNA was extracted from whole blood using chelex-100 and amplified by PCR using the GenePrint STR kit and silver stained. Final DNA profil… Show more

“…Genomic DNA was extracted from oral mucosa exfoliative cell samples with the Chelex-100 method (Manamperi et al 2009). The genetic variations were genotyped with polymerase chain reaction (PCR)-single-strand conformation polymorphism (SSCP), PCR-restriction fragment length polymorphism (RFLP), or amplified fragment length polymorphism methods.…”

“…Other than the applications related to infectious and inherited diseases, DNA fingerprinting to resolve disputed paternity cases and forensic cases is the other strand of molecular based investigations widely being used in Sri Lanka (at present offered only by the Molecular Medicine Unit of the Faculty of Medicine, University of Kelaniya and Genetech Diagnostics, Colombo; [11]). This new technology has had a wide ranging impact on both the civil and criminal branches of the judiciary, and provision of these services has helped to resolve in 1-2 months paternity disputes which have dragged on in some cases for more than 9-10 years, and importantly, to exonerate innocent individuals from criminal charges.…”

The PCR results were visualized using 2% acrylamide gel and then stained using silver staining (21)(22)(23). The primary targets were 123-171 bp for vWA, 322-444 bp for FGA, and 156-195 bp for TH01.…”
