Growth Promotion and Disease Suppression Ability of a Streptomyces sp. CB-75 from Banana Rhizosphere Soil

An actinomycete strain, CB-75, was isolated from the soil of a diseased banana plantation in Hainan, China. Based on phenotypic and molecular characteristics, and 99.93% sequence similarity with Streptomyces spectabilis NBRC 13424 (AB184393), the strain was identified as Streptomyces sp. This strain exhibited broad-spectrum antifungal activity against 11 plant pathogenic fungi. Type I polyketide synthase (PKS-I) and non-ribosomal peptide synthetase (NRPS) were detected, which were indicative of the antifungal compounds that Streptomyces sp. CB-75 could produce. An ethyl acetate extract from the strain exhibited the lowest minimum inhibitory concentration (MIC) against Colletotrichum musae (ATCC 96167) (0.78 μg/ml) and yielded the highest antifungal activity against Colletotrichum gloeosporioides (ATCC 16330) (50.0 μg/ml). Also, spore germination was significantly inhibited by the crude extract. After treatment with the crude extract of Streptomyces sp. CB-75 at the concentration 2 × MIC, the pathogenic fungi showed deformation, shrinkage, collapse, and tortuosity when observed by scanning electron microscopy (SEM). By gas chromatography-mass spectrometry (GC-MS) of the crude extract, 18 chemical constituents were identified; (Z)-13-docosenamide was the major constituent. Pot experiments showed that the incidence of banana seedlings was reduced after using Streptomyces sp. CB-75 treatment. The disease index was 10.23, and the prevention and control effect was 83.12%. Furthermore, Streptomyces sp. CB-75 had a growth-promoting effect on banana plants. The chlorophyll content showed 88.24% improvement, the leaf area, root length, root diameter, plant height, and stem showed 88.24, 90.49, 136.17, 61.78, and 50.98% improvement, respectively, and the shoot fresh weight, root fresh weight, shoot dry weight, and root dry weight showed 82.38, 72.01, 195.33, and 113.33% improvement, respectively, compared with treatment of fermentation broth without Streptomyces sp. CB-75. Thus, Streptomyces sp. CB-75 is an important microbial resource as a biological control against plant pathogenic fungi and for promoting banana growth.

A total of 461 indigenous Streptomycetes strains recovered from various Greek rhizosphere habitats were tested for their bioactivity. All isolates were examined for their ability to suppress the growth of 12 specific target microorganisms. Twenty-six were found to exert antimicrobial activity and were screened for potential nematicidal action. S. monomycini ATHUBA 220, S. colombiensis ATHUBA 438, S. colombiensis ATHUBA 431, and S. youssoufensis ATHUBA 546 were proved to have a nematicidal effect and thus were further sequenced. Batch culture supernatants and solvent extracts were assessed for paralysis on Meloidogyne javanica and Meloidogyne incognita second-stage juveniles (J2). The solvent extracts of S. monomycini ATHUBA 220 and S. colombiensis ATHUBA 438 had the highest paralysis rates, so these Streptomycetes strains were further on tested for nematodes’ biological cycle arrest on two Arabidopsis thaliana plants; the wild type (Col-0) and the katanin mutant fra2, which is susceptible to M. incognita. Interestingly, S. monomycini ATHUBA 220 and S. colombiensis ATHUBA 438 were able to negatively affect the M. incognita biological cycle in Col-0 and fra2 respectively, and increased growth in Col-0 upon M. incognita infection. However, they were ineffective against M. javanica. Fra2 plants were also proved susceptible to M. javanica infestation, with a reduced growth upon treatments with the Streptomyces strains. The nematicidal action and the plant-growth modulating abilities of the selected Streptomycetes strains are discussed.

Introduction Since ancient times medicinal plants have been used as medicine in many parts of the world to promote human health and longevity. In recent years many novel secondary metabolites of plants have been isolated and reported to provide lead compounds for new drug discoveries. Solanum mauritianum Scopoli is native to South America. It is reported to be used by native South Americans during famine as a vegetable and as medicine to cure various diseases. In South Africa the plant is viewed as weed and is facing eradication, however, this plant is a valuable subject for research into its potential pharmaceutical and chemical uses. This study elucidated the metabolic profile of fungal endophytes that have promising bioactive secondary metabolites against pathogenic microorganisms, including mycobacterium species. Material and methods Fungal endophytes from a weed Solanum mauritianum Scop. were used to synthesize secondary metabolites. Gas chromatograph high-resolution time-of-flight mass spectrometry (GC-HRTOF-MS) was used to analyse volatile compounds to prove that potentially fungal endophytes could be extracted from this weed. Extracts obtained with ethyl acetate were screened for phytochemicals and analyzed using a gas chromatograph high-resolution time-of-flight mass spectrometry system. Principal component analysis was used to compare the gas chromatograph high-resolution time-of-flight mass spectrometry data for differences/similarities in their clustering. Phytochemical screening was conducted on the crude extracts of fungal endophytes obtained from different parts of Solanum mauritianum Scopoli (leaves, ripe fruit, unripe fruit and stems). Results Phytochemical screening indicated the presents of alkaloids, flavonoids, glycosides, phenols, quinones and saponins. Quinones were not present in the crude extracts of Fusarium sp. A total of 991 compounds were observed in the fungal endophytes, and Cladosporium sp. (23.8%) had the highest number of compounds, compared to Paracamarosporium leucadendri (1.7%) and Talaromyces sp. (1.5%). Some volatile compounds such as eicosane, 2-pentadecanone, 2-methyloctacosane, hexacosane and tridecanoic acid methyl ester with antibacterial activity were also observed. Conclusion Compositional variations between the plant and fungal endophyte phytochemicals were observed. The results of this study indicate that fungal endophytes from Solanum mauritianum Scop. contain compounds that can be exploited for numerous pharmaceutical and medicinal applications.
