Structural Requirements for Interaction of Peroxisomal Targeting Signal 2 and Its Receptor PEX7

Abstract: Background: Type 2 peroxisomal targeting signals (PTS2) tag proteins for import into peroxisomes. Results: Characterization of structural properties of PTS2 allows the prediction of novel PTS2 and identification of the binding site on the receptor PEX7. Conclusion: PTS2 forms helical structures that bind to a groove on PEX7. Significance: Understanding the recognition of PTS2 by its receptor is a critical step in peroxisomal protein transport.

“…In general, peroxisomal matrix proteins synthesized in the cytosol are bound by peroxisomal receptors – PEX523 in the case of PTS1, and PEX724 in the case of PTS2. These receptor–cargo complexes then move to the peroxisome membrane where they dock with protein sub-complexes that are in or on the membrane.…”

“…Secondary antibodies were goat ␣-mouse-HRP (Dako) and goat ␣-rabbit-HRP (Bio-Rad). All procedures were carried out as described previously (28). Detection was carried out using a Chemidoc XRSϩ detection system (Bio-Rad), and quantitative analysis was performed using Image Lab 4.0 analysis software.…”

“…Although PTS-1 and PTS-2 motif patterns are apparently well conserved in different phyla, correct prediction of proteins with peroxisomal targeting sequences is a challenging task. In silico predictions can be nonetheless done using computational methods; such analyses have been shown to be highly accurate in different systems (Hawkins et al, 2007;Kunze et al, 2011;Lingner et al, 2012;Opperdoes and Szikora, 2006).…”
