Structure and evolution of four POU domain genes expressed in mouse brain.

Abstract: Four mouse POU domain genomic DNA clones-Brain-i, Brain-2, Brain-4, and Scip-and Bran-2 cDNA, which are expressed in adult brain, were cloned and the coding and noncoding regions ofthe genes were sequenced. The amino acid sequences of the four PoU domains are highly conserved; sequences in other regions of the proteins also are conserved but to a lesser extent. The absence of introns from the coding regions of the four POU domain genes and the similarity of amino acid sequences of the corresponding proteins su… Show more

“…The POU domain genes are classi¢ed into six classes [6]. Our interests are in the genes of class III which are mainly expressed in the developing central nervous system, especially in the brain [2,3,7].…”

“…Brain-2 genes have been isolated from di¡erent animals, such as human (BRN-2/N-OCT3/OTF7) [11], mouse (Brain-2) [7] and Xenopus (XLPOU3, XLPOU3b) [12,13]. Functional analyses on transgenic mice and cultured cells show that expression of mouse Brain-2 is required for the development of the endocrine hypothalamus and pituitary gland as well as maturation of speci¢c neurons [8^10,14^16].…”

“…57,58 POUIII transcriptional factors BRN1, BRN2, BRN4 and OCT6 are widely expressed in the developing CNS with extensive regional overlap. Knockdown of the canonical Notch effector C-promoter binding factor 1 (CBF1/RBP-J) promotes the conversion of NSCs to NPs, whereas activation of CBF1 is insufficient to convert NPs back to NSCs.…”
