Abstract:Amide synthases catalyze the formation of macrolactam rings from aniline-containing polyketide-derived seco-acids as found in the important class of ansamycin antibiotics. One of these structurally and functionally hitherto undescribed amide synthases is the geldanamycin amide synthase ShGdmF, which we isolated for the first time and studied in detail both functionally as well as structurally. Here we show that purified ShGdmF catalyzes the amide formation using synthetically derived simplified substrates. The… Show more

scite is a Brooklyn-based organization that helps researchers better discover and understand research articles through Smart Citations–citations that display the context of the citation and describe whether the article provides supporting or contrasting evidence. scite is used by students and researchers from around the world and is funded in part by the National Science Foundation and the National Institute on Drug Abuse of the National Institutes of Health.
