Structure and in vitro activities of a Copper II-chelating anionic peptide from the venom of the scorpion Tityus stigmurus

Abstract: Anionic Peptides are molecules rich in aspartic acid (Asp) and/or glutamic acid (Glu) residues in the primary structure. This work presents, for the first time, structural characterization and biological activity assays of an anionic peptide from the venom of the scorpion Tityus stigmurus, named TanP. The three-dimensional structure of TanP was obtained by computational modeling and refined by molecular dynamic (MD) simulations. Furthermore, we have performed circular dichroism (CD) analysis to predict TanP se… Show more

“…A transcript for a putative anionic HDP without disulfide bridges is also reported. CliHDPAni01 shows similarities to the genome-derived sequence XP_023227050 from C. sculpturatus , and with TanP (GeneBank ID of the transcript: JK483720), a peptide isolated from the venom of Tityus stigmurus [67]. The predicted precursors translated from these sequences, are shown aligned in Figure 5D.…”

“…Its closest match in terms of sequence similarity was Hta1 (55% identity), from a transcriptome analysis of Hadogenes troglodytes . Highly anionic peptides of this kind have been previously found in scorpions [ 55 , 56 ], and in other phyla. They are integral part of the host defense systems of vertebrates, invertebrates and plants [ 57 ].…”

“…The ability to chelate the copper ion from the extracts was determined by the method described by Melo et al [74]. Pyrocatechol violet, the reagent used in this assay, can associate with certain cations, such as aluminum, copper, bismuth and thorium.…”
