Structure of a group A streptococcal phage-encoded virulence factor reveals a catalytically active triple-stranded β-helix

Abstract: Streptococcus pyogenes (group A Streptococcus) causes severe invasive infections including scarlet fever, pharyngitis (streptococcal sore throat), skin infections, necrotizing fasciitis (flesh-eating disease), septicemia, erysipelas, cellulitis, acute rheumatic fever, and toxic shock. The conversion from nonpathogenic to toxigenic strains of S. pyogenes is frequently mediated by bacteriophage infection. One of the key bacteriophage-encoded virulence factors is a putative ''hyaluronidase,'' HylP1, a phage tail-… Show more

“…Comparable structures include the triple β-helix regions of phage T4 gp12 (41) and gp5 (42), the K1F endosialidase (43) and endo-N-acetylneuramidase (29), streptococcal (pro)phage HylP1 and Hylp2 (44,45), the phage P22 cell penetrating needle gp26 (40), and phage Phi29 tail-spike (46). However, the most similar structures are those of phage P2 gpV (Fig.…”

“…Recently, a GH69 enzyme was found to be a lyase, and reassigned to a polysaccharide lyase family, PL16. 60,61) Phosphorylases have been assigned EC numbers of glycosyltransferases (2.4.1. ) according to the apparent reaction scheme.…”

“…Crystal structures of lyases belonging to families PL-1, -3, -4, -5, -6, -7, -8, -9, -10, -16, and -18 have been determined. These enzymes are grouped into six structural categories: (i) the parallel ␤-helix in PL-1 (13,14,16,17,55,56), -3 (57), -6 (58), and -9 (20); (ii) the ␣/␣-barrel in PL-5 (59) and -10 (60, 61); (iii) the ␣/␣-barrel plus anti-parallel ␤-sheet in PL-8 (29,(62)(63)(64)(65)(66); (iv) the ␤-sandwich plus ␤-sheet in PL-4 (33); (v) the ␤-jelly roll in PL-7 (27,67,68) and -18; and (vi) the triplestrand ␤-helix in PL-16 (69). The folds and substrates of polysaccharide lyases are summarized in Table 1.…”
