Structure of cDNAs Encoding Human Eukaryotic Initiation Factor 3 Subunits

Abstract: The mammalian translation initiation factor 3 (eIF3), is a multiprotein complex of ϳ600 kDa that binds to the 40 S ribosome and promotes the binding of methionyltRNA i and mRNA. cDNAs encoding 5 of the 10 subunits, namely eIF3-p170, -p116, -p110, -p48, and -p36, have been isolated previously. Here we report the cloning and characterization of human cDNAs encoding the major RNA binding subunit, eIF3-p66, and two additional subunits, eIF3-p47 and eIF3-p40. Each of these proteins is present in immunoprecipitates … Show more

“…Jab1 belongs to the Mov-34 protein family whose members are involved in diverse functions involving macromolecular assembly (Asano et al, 1997). Jab1 has been originally identi®ed by its ability to bind to the c-Jun transactivation domain (Claret et al, 1996).…”

“…To narrow the TRC8 interaction domain in JAB1, we generated labeled proteins for amino acids 1 ± 187 containing the Nterminus and MPN protein interaction domain, the MPN domain alone (aa 55 ± 187), and the C-ter (aa 191 ± 327). Both proteins containing the MPN domain (Asano et al, 1997;Hofmann and Bucher, 1998;Ferrell et al, 2000) bound DTrc8 while the C-ter did not (Figure 4d, lower panels).…”

“…The EIF3S3 gene encodes for a p40 subunit of eukaryotic translation initiation factor 3 (eIF3). The eIF3 complex plays a central role in the translation initiation pathway by binding to 40S ribosomal subunits in the absence of other translational components, and it helps to maintain the 40S and 60S subunits in a dissociated state (Asano et al, 1997). The function of the p40 subunit itself is, however, not known.…”
