Structure of the 16 S ribosomal RNA of the thermophilic cyanobacterium chlorogloeopsis HTF (‘mastigocladus laminosus HTF’) strain PCC7518, and phylogenetic analysis

Abstract: The thermophilic cyanobacterial strain, PCC7518, originally identified as 'Mu~tigocludus hminosus HTF' does not show branchings or heterocysts. The absence of branchings supports the later assignment to the genus Chlorogloeopsu. The absence of heterocysts may be the result of a mutation because heterocysts were observed in the original isolate. Alternatively, contamination may have happened. To solve this problem, the 16 S rRNA sequence was determined and used to infer a secondary structure model and build dis… Show more

“…The validity of a gap between Mastigocladus and unbranched Chlorogloeopsis (KOMÁ-REK and HAUER 2010) was proved also by 16S rRNA analysis (WILMOTTE et al 1993); they also proposed that the ability to produce heterocytes can be lost by mutation. Also, Chlorogloeopsis fritschii is not so homogenous a species, since two different types of germination of akinetes were observed (HINDÁK 2008).…”

“…16S rRNA gene libraries of 13 C-enriched DNA 16S rRNA clone libraries, each comprising 96 clones, were prepared from combined fractions containing the 13 C-enriched DNA from each of the three SIP incubations using general eubacterial primers 27f and 1492r and partially sequenced using primer 27f (Wilmotte et al, 1993) at the Beckman Coulter Genomics sequencing facility (Danvers, MA, USA). The 13 C-enriched heavy DNA fractions were selected based on the DGGE evidence, which is discussed below.…”
