Structures and Cytotoxic Evaluation of New and Known Acyclic Ene-Ynes from an American Samoa Petrosia sp. Sponge

Abstract: Four new compounds, (−)-petrosynoic acids A–D (1–4), and five known congeners, pellynols A (5), C (6), D (7), F (8), and I (9) were isolated from a Petrosia sp. marine sponge collected in American Samoa. Isolation work was guided by cytotoxicity against human lung cancer cells (H460). The structures of the C31-C33 polyacetylenes (1–9) were determined on the basis of 1D- and 2D-NMR analysis, mass spectrometry, and comparison of specific rotation values. Compounds 1–9 were found to be broadly cytotoxic with limi… Show more

“…607 Petrosynic acids A-D 567-570 (Petrosia sp., Tutuila, American Samoa) all displayed similar activity versus various HTCLs and non-proliferative human broblasts and hence no therapeutic window is available. 608 A New Caledonian Niphates sp. was the source of nepheliosyne B 571.…”

“…The analytical LC-UV-ELSD-MS system was controlled by the Empower software and comprised Waters HPLC components equipped with a reversed-phase column (Phenomenex, Luna C 18 , 150 × 4.6 mm, 5 μm) and ran at 1.0 mL/min. The operating method was the same with our previous report [22]. The semi-preparative HPLC system comprised Waters HPLC components (a gradient controller, two 515 pumps and one 2487 UV-Visible detector) and was equipped with a reversed-phase semi-preparative column (Phenomenex, Synergi Hydro-RP, 250 × 10.0 mm, 4 μm).…”
