StuA is required for cell pattern formation in Aspergillus.

Abstract: The stunted (stuA) gene product is required for the orderly differentiation and spatial organization of cell types of the Aspergillus nidulans conidiophore. Expression of the stuA gene is complex. Two transcripts, stuA~ and stuA~, are initiated from separate promoters. Transcription of both RNAs increases -50-fold during the establishment of developmental competence. Induction-dependent transcriptional and post-transcriptional regulatory mechanisms further enhance expression 15-fold. Consistent with the latter… Show more

“…nidulans For the targeting of GFP to nuclei, the A. nidulans StuA protein was used. The stuA gene encodes a putative transcription factor that is involved in the regulation of conidiophore development (Miller et al, 1992). At the carboxyterminus of the protein, a putative bipartite nuclear localization domain was detected.…”

“…Several key regulatory genes have been identified and cloned and shown to control progression through the various stages of conidiation (28)(29)(30)(31). Properties of the corresponding regulatory proteins and the sites at which some of them act have been determined (32,33).…”

“…Several hyphal inducing media were used: Lee's medium (36) with 1% mannitol as the carbon source, YPD ϩ 10% serum, SS (synthetic succinate) medium (0.0425% YNB without amino acids and ammonium sulfate (Difco), 0.125% ammonium sulfate, 2% succinic acid, pH 6.5), and SSA (SS with amino acids) medium. A modified lithium acetate method was used for C. albicans transformation (34).…”

“…The fact that AbaA expression is under the direct control of StuA in A. nidulans (36), and that AbaA/Tec1-binding sites are found in the upstream regions of all known hypha-specific and serum-induced genes (Table IV), led us to investigate the possibility that Efg1 may regulate TEC1 transcription. We examined TEC1 expression in efg1/efg1 mutants by Northern analysis.…”
