Studies on correlations between soil chemistry and bacterial population in rhizosphere of Bt and non-Bt cotton and characterization of rhizobacteria

Abstract: The present study was planned to explore the relationship between soil chemistry with bacterial population of Bt and non-Bt cotton and their phenotypic and molecular characteristics. The pre-plant soil and rhizospheres of Bt and non-Bt cotton were collected and analyzed for some soil parameters and bacterial population. The bacterial isolates were analyzed for their morphological, biochemical, and molecular characteristics. Bacterial population showed a significant (p < 0.05) positive correlation with clay con… Show more

“…The increase in EC with Rw can be attributed to the quantity of negatively charged ions in the soil that interact with soluble salts. Due to the increased acidity, soil EC rose with the BMA when organic acid is released during decomposition (Ibrahim et al, 2020). The chisel ploughing with Rc caused a short-term decrease in soil pH at sowing compared to the cultivator in 2018 and 2019, which persisted until maize harvest.…”

“…The deoxyribonucleic acid (DNA) of bacterial isolates producing pink and yellow-colored colonies in Luria-Bertani broth culture was extracted by previously reported method using commercially available DNA extraction kit (Vivantis kit; catalog: GF-BA-100 preps, USA) (Ibrahim et al 2020). The ampli cation of bacterial speci c 16S ribosomal RNA gene was done using universal primer derived from 16S rRNA sequence of Escherichia coli in a thermal cycler polymerase chain reactor (Agilent, Sure Cycler 8800, USA) (Porteous and Armstrong, 1993).…”

“…The ampli cation was carried out using universal primers: forward primer 5'CAGCAGCCGCGGTAATAC3' and reverse primer 5'ACGGGCGGTGTGTACAAG3'. The process of ampli cation was completed following the experimental protocols as reported by Ibrahim et al 2020 (Ibrahim et al 2020).…”
