Study on an antimicrobial protein produced by Paenibacillus polymyxa JSa-9 isolated from soil

Abstract: Paenibacillus polymyxa strain JSa-9, a soil isolate showed in vitro inhibitory effects on several pathogens. A protein of about 71.9 kDa, isolated from a culture of JSa-9, exhibited broad-spectrum antibacterial and antifungal activities. The purification procedure consisted of ion-exchange chromatography on DEAE-Sephacel and Sephadex G-100 column chromatography. The purified protein was characterized to be a glycoprotein, which contained 53% of amino acids and 28% of carbohydrate. Its N-terminal sequence was d… Show more

“…Among these, Paenibacillus populations were found to be capable of suppressing several plant diseases and promoting plant growth [5, 6]. Paenibacillus populations synthesize the plant hormones auxin and cytokinin, solubilize soil phosphorus, and produce biological control substances like siderophores and antibiotics [7].…”

“…Serial dilutions of antimicrobial compounds were prepared to obtain final concentrations of 1000, 500, 250, 125, 62.5, 31.25, 15.63, and 7.81 μg/ml in LB or PDA medium. The ability of cyclic products strains to inhibit the growth of various indicator organisms was determined qualitatively by agar-well diffusion method (Deng et al, 2011a). Plates were incubated for up to 24 (bacteria) and 48 h (fungi) before the antimicrobial activities were determined (Oh et al, 2011).…”

“…Liu et al ( 2007 ) used a DEAE-Sepharose Fast Flow weak ion exchange column to chromatograph the crude protein secreted by B. subtilis B-916 strain, and after obtaining an active elution peak, the protein spectrum identified a novel antibacterial protein, Bacisubin, with a molecular weight of 41.900 kDa. The purification procedure of ion-exchange chromatography on DEAE-Sephacel and Sephadex G-100 column chromatography isolated a glycoprotein with a molecular weight of approximately 71.9 kDa from Paenibacillus polymyxa Jsa-9 fermentation broth, which exhibited broad-spectrum antibacterial and antifungal activities (Deng et al 2011 ).…”
