Study on the interaction of anti‐inflammatory drugs with human serum albumin using molecular docking, quantitative structure–activity relationship, and fluorescence spectroscopy

Abstract: The interaction of 14 anti‐inflammatory drugs with human serum albumin (HSA) was investigated using fluorescence quenching, molecular docking studies, and quantitative structure–activity relationship (QSAR) methodology. Binding of anti‐inflammatory drugs to HSA plays a fundamental role in their transport, distribution, delivery, and elimination. Binding constants of these drugs to HSA, obtained using the fluorescence quenching method, were within the range 0.01 × 104 M−1 (acetaminophen) to 1881.05 × 104 M−1 (m… Show more

“…The results listed in Table 4 made visible that the binding constants indicate values of about 10 5 dm 3 ·mol −1 at 297 K and decrease in higher temperatures. Similar values were obtained for many biologically active compounds [ 44 , 47 , 48 , 49 , 50 , 51 ]. Thus, the number of binding sites is close to 1 for all of the studied compounds, which means a one-to-one interaction.…”

“…In general, static quenching is a ground state phenomenon that occurs due to the formation of a complex between fluorophore (DNA/BSA) and quencher 4g in the ground state, whereas; dynamic quenching is an exciting state phenomenon that arises due to collisions between fluorophore and quencher in the excited state 37 . How quenching proceeds can be easily identified by calculating the quenching constant Kq using the Stern–Volmer equation 38 (Eq. 1 ).…”

“…The interaction of 14 anti-inflammatory drugs with human serum albumin was investigated by F. Mohammadnia [ 32 ]. The binding constants were found with the range 10 2 dm 3 ·mol −1 (acetaminophen) to 1.88 × 10 7 dm 3 ·mol −1 for meloxicam.…”

“…However, in the literature, there are several models to determine the binding parameters. The values obtained from different methods of calculation could significantly differ from each other [30][31][32]. Each of them also has various limitations.…”
