Subcellular-level resolution MALDI-MS imaging of maize leaf metabolites by MALDI-linear ion trap-Orbitrap mass spectrometer

Abstract: A significant limiting factor in achieving high spatial resolution for matrix-assisted laser desorption ionizationmass spectrometry (MALDI-MS) imaging is the size of the laser spot at the sample surface. Here, we present modifications to the beam-delivery optics of a commercial MALDI-linear ion trap-Orbitrap instrument, incorporating an external Nd:YAG laser, beam-shaping optics, and an aspheric focusing lens, to reduce the minimum laser spot size from ~50 μm for the commercial configuration down to ~9 μm for … Show more

“…MALDI-MSI can obtain high-spatial-resolution images of ions, allowing for the visualization of fine structures. Spatial resolution of ;20 mm has become routine in MALDI-MSI, and 2-to 5-mm spatial resolution has been demonstrated by several groups, with the latter providing a means to locate metabolites within subcellular compartments (Zavalin et al, 2013;Korte et al, 2015).…”

“…Procedures for corn seed germination and plant growth are described in detail elsewhere [11,12], and briefly described below. A maize seed was imbibed for 10 min and placed with the embryo side facing down on top of moist filter paper in a Petri dish that was placed in a climate-controlled greenhouse.…”

“…The instrument was modified to use an external 355 nm Nd:YAG laser (UVFQ; Elforlight Ltd., Daventry, UK) and a custom optical set-up [12]. TunePlus and Xcalibur software (Thermo Scientific) were used to define imaging acquisition parameters and for data acquisition.…”

“…The technique enables simultaneous label-free visualization of the lateral distribution of numerous biomolecules in the sections (e.g., peptides, lipids, further metabolites). With current MALDI-MSI instruments, lateral resolutions in the range of a few 10 μm are routinely achieved, and several reports even demonstrated MALDI-MSI analysis with pixel sizes in the low micrometer range [3][4][5][6][7].…”
