Succinate Dehydrogenase Subunit C Contributes to Mycelial Growth and Development, Stress Response, and Virulence in the Insect Parasitic Fungus Beauveria bassiana

Abstract: The electron transport chain (ETC) is critical for energy supply by mediating the electron flow along the mitochondrial membrane. Succinate dehydrogenase (SDH) is also known as complex II in the ETC, in which SdhC is a subunit anchored in mitochondrial membrane. However, the physiological roles of SdhC remain enigmatic in filamentous fungi.

“…Previous studies have indicated that SdhC was essential in maintaining SQR function and SdhC deletion mutants were either unsuccessful to obtained or resulted in mutants with a significant decrease in fitness. , To validate the function of FgSdhC and FgSdhC 1 , targeted deletions of two genes were performed in the isolates PH-1, F1405, and Fg131 (Figure S3). PH-1 and Fg131 were F.…”

“…Similarly, in Beauveria bassiana, two SdhC species ( BbSdhC 1 and BbSdhC 2 ) were identified. BbSdhC 1 played a role in maintaining SQR function but that BbSdhC 2 was not sufficient …”

“…41 Loss of the BbSdhC 1 in Beauveria bassiana leads to heightened intracellular ROS levels and impaired mycelial growth under oxidative stress conditions. 42 Mutations in SdhC of embryonic fibroblasts have been shown to increase ROS production and enhance tumorigenesis in human cells. 43,44 SDH also plays a significant role in plants regarding ROS production, regulation of plant development, and stress responses.…”

“…The amplified fragments and the linearized pRS426 plasmid were cotransformed into S. cerevisiae (FY834) to obtain the correct knockout vector pRS426-AoSte12-hph, and the disruption sequences were amplified from it using Ste12LF1 and Ste12RR1 and then transformed into A. oligospora protoplasts, as previously described ( 67 , 68 ). The positive transformants were confirmed via PCR and Southern blotting, as described previously ( 45 , 69 ).…”
