Supramolecular Radiosensitizer Based on Hypoxia‐Responsive Macrocycle

Abstract: Radiotherapy (RT) has been viewed as one of the most effective and extensively applied curatives in clinical cancer therapy. However, the radioresistance of tumor severely discounts the radiotherapy outcomes. Here, an innovative supramolecular radiotherapy strategy, based on the complexation of a hypoxia-responsive macrocycle with small-molecule radiosensitizer, is reported. To exemplify this tactic, a carboxylated azocalix[4]arene (CAC4A) is devised as molecular container to quantitatively package tumor sensi… Show more

“…We determined that the fluorescence intensity of NAQA⊂Zn-MPPB was considerable weak in untreated A549 and MCF- control), because of the relatively low content of intracellular NTR. In contrast, under the hypoxia conditions (8% and 0.1% O 2 ), an obvious fluorescence signal corresponding to NAQA⊂Zn-MPPB 54 was observed (Figure 4A). As the hypoxia degree increased, the effect of cell fluorescence imaging becomes more obvious, over 3-fold stronger fluorescence emission intensity of MCF-7 cells in 0.1% O 2 than under normoxic conditions (see Figure S43A in the Supporting Information).…”

“…6 ). Calixarenes were selected as the macrocyclic scaffold because of their extensive chemical design space 50 53 . By directly reacting calix[4]arene with p -carboxybenzenediazonium chloride at 2 °C for 3 h, CAC4A, which can be linked by condensation reaction with propargyl amine, getting CAC4A-Alk, was obtained with quantitative yield.…”
