Susceptibility to Anthrax Lethal Toxin-Induced Rat Death Is Controlled by a Single Chromosome 10 Locus That Includes rNlrp1

Abstract: Anthrax lethal toxin (LT) is a bipartite protease-containing toxin and a key virulence determinant of Bacillus anthracis. In mice, LT causes the rapid lysis of macrophages isolated from certain inbred strains, but the correlation between murine macrophage sensitivity and mouse strain susceptibility to toxin challenge is poor. In rats, LT induces a rapid death in as little as 37 minutes through unknown mechanisms. We used a recombinant inbred (RI) rat panel of 19 strains generated from LT-sensitive and LT-resis… Show more

“…However, the results from the experiment comparing lethal toxin in SpragueDawley and Lewis hearts (experiment 7) do not support such a relationship. Sensitivity to lethal toxin-induced death in rats has been mapped to a single locus, rNlrp1 (23). While variations in this genetic locus conferred differing survival benefits with lethal toxin in vivo, in experiment 7 rNlrp1 did not appear to differentially influence myocardial function.…”

“…To examine whether the resistance to the lethal effects of lethal toxin observed in vivo in Lewis rats (23) was reproducible in an isolated heart model, hearts from SpragueDawley and Lewis rats were perfused in a nonrecirculating model Values are means Ϯ SE. *Controls included animals receiving diluents of edema toxin or PA alone.…”

“…There are two NLRP1 paralogs in rats (Nlrp1a and Nlrp1b) (24): the Nlrp1a paralog is associated with rat macrophage susceptibility to LeTx (25), but little is known about the Nlrp1b paralog. Amino-terminal cleavage of murine NLRP1B or rat NLRP1A in response to Toxoplasma gondii has not been demonstrated, and it might be that the mechanism of activation is distinct from that of LeTx.…”
