Sustained-release alginate-chitosan pellets prepared by melt pelletization technique

Abstract: Similar to calcium alginate pellets, alginate-chitosan pellets demonstrated sustained drug release property though via different mechanisms. Combination of alginate, chitosan and calcium acetate in the same matrix nevertheless failed to retard drug release via complementary drug release pattern.

“…The solution viscosity of the modified chitosan was determined using an already reported method [47]. Briefly, a 0.5% (w/w) solution of each chitosan sample was prepared in 2% (v/v) acetic acid and filled into a U-tube viscometer (Ostwalds Viscometer, Zhejiang, China).…”

“…The calcium content of melt pellets undergoing drug dissolution was examined using atomic absorption spectrophotometry technique (Z-2000, Hitachi Hi-Technologies Corporation, Japan), adopting a previously reported protocol (Wong and Nurulaini, 2012). It was calculated from the difference between total calcium content of pellets and amount of calcium released in dissolution medium at specific intervals.…”

“…The dispersed fragments, though exhibiting a larger specific surface area for drug dissolution than intact matrix, are rapidly crosslinked and consolidated by soluble Ca 2+ from calcium acetate and have drug release retarded. In another study of sustained-release alginate pellets prepared by melt pelletization technique, the chitosan instead of calcium acetate is used to coacervate with alginate into polyelectrolyte complexes (Wong and Nurulaini, 2012). The slow drug release characteristics are a resultant effect of pellet aggregation into swollen structures with reduced erosion.…”

“…Chitosan is a linear randomly distributed, binary heteropolysaccharide consisting of β(1‐4) linked 2‐acetamido‐2‐deoxy‐β‐D‐glucopyranose and 2‐amino‐2‐deoxy‐β‐D‐glycopyranose units (Wong, ; Wong & Harjoh, ; Yong & Wong, ). It is a natural, biodegradable and biocompatible polymer which lacks irritant and allergic effects.…”
