Synergic effect between TRAIL gene and curcumin in magnetic chitosan nanoparticles on cancer cells apoptosis enhanced by laser photoactivation

Abstract: The Apoptosis is an intensely studied mechanism for cancer therapy. Various genes, drugs, and molecules have been combined to potentiate the apoptotic effect against tumor cells. Magnetic nanocomplexes with biopolymers have been proposed as nanocarriers for delivery of active molecules and improve apoptosis induction. To these purpose were formulated magnetic nanoparticles with curcumin coated chitosan (MNPs-CHI-TPP-CUR) and magnetic nanoparticles with pCEM-TRAIL plasmid coated chitosan (MNPs-CHI-TPP-TRAIL) fo… Show more

“…To prove the efficiency of our vector, it was encapsulated in chitosan magnetic nanoparticles, called Magnetogene, with a final size of 60 nm by SEM and 218 nm by DLS ( Figure 2 ). DLS requires the sample to be dispersed in solution, while for SEM, a dry flat surface sample is used, and the difference in the sample can influence the morphology of the Magnetogene due to the flexible nature of the chitosan polymer [ 20 , 28 ]. Magnetogene has a Zeta potential of +15.9 mV which is a critical parameter for cellular interaction; interactions with pDNA-NP complexes indicate that the complexes were positively charged.…”

“…Among these polymers, chitosan is an attractive drug or gene delivery system [ 17 , 18 ]. Magnetic drug delivery systems work via the delivery of magnetic nanoparticles loaded with the drug to the tumor site under the influence of an external magnetic field [ 19 , 20 ]. In vivo magnetic fields are focused on the required site to promote transfection and to deliver the therapeutic gene to a specific organ or site within the body.…”

“…This was done either alone or in combination with the aim of overexpressing the receptors required for apoptosis induction. Results revealed a significant increase in cellular death within 48 h when using a combination of TRAIL gene-based and drug-based systems compared to each nanocomplex individually, indicating a synergistic effect [127].…”

“…Curcumin (Cur) and chitosan (Chi) encapsulation within MNP was performed according to previous reports (22,23). Water-soluble Chi (20 kDa) was purchased from Coyote Foods Biopolymer and Biotechnology (Saltillo, Coahuila, Mexico) and resuspended in MilliQ water at a concentration of 2 mg/mL.…”
